Membrane lipid modification of chinese hamster ovary cells. Thermal properties of membrane phospholipids.

چکیده
A simple and reproducible method for altering the acyl chain composition of Chinese hamster ovary cell phospholipids is described. By supplementation of the medium with 5% delipidized serum and 12 pg/ml of various fatty acids, extensive incorporation of the added fatty acid into membrane phospholipids was obtained. No significant changes in the phospholipid head group composition or the sterol/phospholipid ratio were detected during the experimental time course. Analysis of the thermal properties of these supplemented phospholipids is also described. The onset of formation of a “solid” lipid phase was detected using the fluorescent fatty acid probe, trans-parinaric acid. The temperature of onset of the solid lipid phase in extracted phospholipids was correlated with the percentage of unsaturated fatty acids esterified to the phospholipids. Fluorescence polarization measurements with cisand trans-parinaric acid in the phospholipids indicated that the amount of solid phase lipid was significantly greater in palmitate-supplemented cells than in unsupplemented or oleate-supplemented cells. The polarization analysis also indicated that the amount of solid phase lipid was approximately equal in unsupplemented and oleate-supplemented cells. Analysis of purified phosphatidylcholine and phosphatidylethanolamine from oleateand palmitate-supplemented cells indicated that the onset of solid formation in these phospholipids occurred at approximately the same temperature as the onset in the total phospholipid extract.

Alteration of the Membrane Lipid Composition and Infectivity of Vesicular Stomatitis Virus
The cholesterol and phospholipid composition of the membrane of vesicular stomatitis (VS) virus was altered by growth in a sterol auxotroph Chinese hamster ovary (CHO MI) host cell and by infection of CHO MI and baby hamster kidney (BHK)-%l cells supplemented with fatty acids and dimethylethanolamine. VS virus released from infected CHO MI sterol auxotroph cells grown in delipidated serum had a...
متن کاملDiacylglycerol generated in CHO cell plasma membrane by phospholipase C is used for triacylglycerol synthesis.
The diacylglycerol (DAG) signal generated from membrane phospholipids by hormone-activated phospholipases is attenuated by mechanisms that include lipolysis or phospholipid resynthesis. To determine whether the DAG signal might also be terminated by incorporation of DAG into triacylglycerol (TAG), we studied the direct formation of TAG from endogenous DAG generated by bacterial phospholipase C ...
متن کاملDetermination of plasma membrane lipid mass and composition in cultured Chinese hamster ovary cells using high gradient magnetic affinity chromatography.
We have utilized wheat germ agglutinin conjugated to iron/dextran particles in conjunction with high gradient magnetic affinity chromatography (HIMAC) to prepare plasma membranes from cultured cells. Membrane-impermeable succinimidyl esters inactivate alkaline phosphodiesterase 1 (APDE-1) and were used to establish the proportion of APDE-1 expressed at the cell surface. The yield of inhibitable...
متن کاملRegulation of phospholipid scramblase activity during apoptosis and cell activation by protein kinase Cdelta.
Phospholipid scramblase induces nonspecific bidirectional movement of phospholipids across the membrane during cell activation and has been proposed to mediate the appearance of phosphatidylserine (PS) in the plasma membrane outer leaflet during apoptosis, a cell surface change that is critical for apoptotic cell removal. We report here that protein kinase C (PKC) delta plays an important role ...
متن کاملSingle cell 3-D platform to study ligand mobility in cell-cell contact.
Lateral mobility and dimensionality have both been shown to influence cellular behavior, but have yet to be combined and applied in a single in vitro platform to address, e.g., cell adhesion in a setting mimicking the three-dimensional environment of neighboring cells in a reductionist way. To study the effect of the lateral mobility of cell adhesive ligands in three dimensions we present and c...
